目的:血管生成抑制剂靶向治疗肿瘤已成为目前研究热点之一,本研究旨在观察苏拉明(Suramin)对肺腺癌小鼠异体移植瘤生长和转移的抑制作用,并初步探讨苏拉明抑制肿瘤生长和转移的相关机制。方法:构建肺腺癌高转移小鼠移植瘤模型,将24只接种Lewis肺癌的C57BL/6J小鼠按随机数字表法随机分成三组:对照组、顺铂组、苏拉明组。给予相应药物干预,于接种后24日处死各组小鼠,经眼球静脉丛取各组小鼠血液样本1 ml,酶联免疫吸附法(Enzyme-linked immunosorbent assay,ELISA)测定VEGF及MMP-9的表达,剥离皮下肿瘤,称重,计算抑瘤率,剖胸取出双肺,计数肺表面转移结节,计算肺转移抑制率。结果:顺铂组、苏拉明组的抑瘤率分别为3920、4911,对照组、顺铂组、苏拉明组的肺表面转移结节数分别为(1400±290)、(977±165)、(600±170),血清VEGF的浓度分别为(43115±2738)pg/ml、(40196±2551)pg/ml、(13535±1905)pg/ml,MMP-9的浓度分别为(22347±2150)ng/ml、(20812±1729)ng/ml、(8902±751)ng/ml。结论:VEGF及MMP-9的高表达促进肺腺癌的生长和转移,苏拉明对肺腺癌移植瘤的生长和转移具有抑制作用,其机制与其抑制VEGF及MMP-9的表达,抑制微血管形成,促进肿瘤细胞侵袭相关。 ObjectiveThe scheme of tumor therapy through angiogenesis becomes one of studying hot spots at present The experiment is designed to observe that if suramin has synergetic effects of suppressing lung adenocarcinoma’s growth,metastasis and to study the related mechanism of suramin’s suppressing tumor growth Methods24 C57BL/6J mice bearing Lewis cells were randomized into 3 groups:control group; DDP group; suramin group; Each group to be the appropriate intervention,all the mice were sacrificed by tumor inoculation in 24 days,Dissectted of subcutaneous tumor and removed of lung,The metastatic tumor foci on lung surface and subcutaneous tumor in mice were weight,tumor inhibitory rate were counted,lung wet weight were observed,lung metastatic occurring rate and metastatic foci inhibitory rate were counted;serum VEGF and MMP-9 were determined by enzyme-linked immunosorbent assayResultscisplatin group,suramin group of tumor growth inhibitory rate were 3920,4911; Control group,cisplatin group,suramin group of lung metastatic nodules were 1400±290、977±165、600±170,the expression of serum VEGF were 43115±2738 pg/ml,40196±2551 pg/ml,13535±1905 pg/ml; the expression of serum MMP-9 were 22347±2150 ng/ml,20812±1729 ng/ml,8902±751 ng/mlConclusionThe high expression of VEGF and MMP-9 promotes growth and metastasis of lung adenocarcinoma,suramin can produce a synergetic inhibitory activity against tumor growth and metastasis of lung adenocarcinomaThe fact that suramin inhibited tumor growth and metastasis of lung adenocarcinoma may be associated its effect of suppressing the expression of VEGF and MMP-9 in mice
